to visualize the temperature of the sample (transparent spheres), the solid-liquid interfaces (transparent lines), and the isotherms, we created a heat transfer model (fig 1). we plan to use this chip for future drug analysis work and use this work to prototype a custom drug.monitoring system. we will also create a 3d-heat transfer model to visualize the thermal gradients across a 50 microliter chip. theoretical and experimental analyses will be performed to verify and optimize the application for drug analysis.

to study the thermal gradients across the measurement region in channels a through c in a 50 microliter chip (approx. 9c to 28c), we used the same techniques and equations as in the proof-of-concept measurements to the 50 microliter chip. in the fig 4b image below, the interface locations are blue, the interface locations are red, and the material boundaries are white. as in the proof-of-concept, we imported the entire measurement region into our software and clicked on the location of each solid-liquid interface. the micrograph in fig 4b shows how the solid-liquid interfaces are located in different locations along the chip (channel a near the bottom, channel b near the bottom, channel c near the middle, and channel d near the bottom). after importing this image into our software, we clicked on each solid-liquid interface, plotted the locations of each interface along the channels, and fit a linear temperature gradient vs. vertical location of the solid-liquid interface using linear regression (fig 4b inset).

in any case, we found the microfluidic thermometer chip to be a stable and accurate method for accurately measuring temperatures within the measurement region (the area of the chip where the temperature gradient is linear; dotted boxes in fig 2a and 2b). in a single study, we used our microfluidic thermometer chip to measure the temperature within all five microfluidic channels simultaneously; our results are shown in fig 2c. figs 2d and 2e show the temperature measurements of a single microfluidic thermometer channel, and fig 2f shows the results of the microfluidic thermometer chip as a whole. the microfluidic thermometer chip is tolerant of significant differences in the temperature of the measurement region, and is not affected by temperature fluctuations in the surrounding environment.
in addition to measuring temperatures, the microfluidic thermometer chip can also be used as a heater in the measurement region (the area of the chip where the temperature gradient is linear; dotted boxes in fig 2a and 2b). the thermoelectric cooler is turned off (i.e., the polarity is reversed) and the microfluidic thermometer chip used as a heat source to change the temperature of a specific fluid sample from 2c to 55c as described in fig 2g. however, its current design is not capable of measuring thermal gradients below 0c because the solidification/melting point of the material in the channels is a few degrees higher than the lowest measured temperature.
